Genetic diversity of mole cricket Gryllotalpa orientalis Burmeister, 1838 in Roi Et Province

Abstract

The mole cricket is an edible, medicinal, and agriculturally significant insect that can be found worldwide. In Thailand, only Gryllotalpa orientalis has been recognized. However, a comprehensive investigation on the genetic variation of mole cricket in Thailand is still lacking. Thus, this study aims to examine the genetic variation of mole cricket from different localities in Roi Et Province. The 271 mole crickets were collected from natural habitats in 14 different districts of Roi Et Province for genetic variation investigation using two mitochondrial genes, namely 430 bp of cytochrome c oxidase subunit 1 (CO1) and 465 bp of 16S ribosomal DNA (16S rDNA) as genetic markers. A high level of genetic variation was found in mole cricket populations in this study. The 97 (22.6%) and 80 (17.2%) variation sites were found in CO1 and 16S rDNA sequences, respectively. According to these variations, 37 (Go1 – Go37) and 15 (Gr1 – Gr15) haplotypes were classified based on CO1 and 16S rDNA sequences, respectively. Haplotype network analysis revealed that six genetic groups have been classified, namely genetic groups I – VI. Phylogenetic tree analysis also demonstrates the six genetic groups, similar to haplotype network analysis. The p-distance value compared between 14 populations ranged from 0.0002 to 0.1138. In addition, morphometric analysis also supports the classification of genetic groups, as demonstrated that some characters showed significant differences between genetic groups, such as thorax width, tegmen length, wing length, and wing width. Thus, mole cricket populations in Roi Et Province contained a species complex. Further comprehensive investigations on biology, ecology, morphology, and genetics need to be examined for a better understanding of their systematic and taxonomic status.

แมลงกระชอนเป็นแมลงที่มีความสำคัญทั้งทางด้านอาหาร การเกษตร และการแพทย์ สามารถพบได้ทั่วโลก แมลงกระชอนในประเทศไทยถูกจัดจำแนกเป็นชนิดเดียว ได้แก่ Gryllotalpa orientalis แต่อย่างไรก็ตาม ยังไม่มีข้อมูลการศึกษาเชิงลึกทางด้านความหลากหลายทางพันธุกรรมของแมลงกระชอนในประเทศไทยมาก่อน ผู้วิจัยจึงสนใจศึกษาความหลากหลายทางพันธุกรรมของแมลงกระชอนที่กระจายอยู่ในพื้นที่จังหวัดร้อยเอ็ด โดยทำการเก็บตัวอย่างแมลงกระชอนจำนวน 271 ตัวอย่าง จากแหล่งธรรมชาติใน 14 พื้นที่แต่ละอำเภอของจังหวัดร้อยเอ็ด มาทำการวิเคราะห์ความหลากหลายทางพันธุกรรมโดยใช้ยีน cytochrome c oxidase subunit 1 (CO1) ความยาว 430 คู่เบส และ 16S ribosomal DNA (16S rDNA) ความยาว 465 คู่เบส เป็นเครื่องหมายโมเลกุล ผลการศึกษาพบว่า ประชากรแมลงกระชอนจากแหล่งธรรมชาติในจังหวัดร้อยเอ็ดมีความหลากหลายทางพันธุกรรมค่อนข้างสูง พบการแปรผันของลำดับนิวคลีโอไทด์ของยีน CO1 และ 16S rDNA จำนวน 97 (22.6%) และ 80 (17.2%) ตำแหน่ง สามารถจำแนกลักษณะทางพันธุกรรมออกได้เป็น 37 (Go1 – Go37) และ 15 (Gr1 – Gr15) แฮโพลไทป์ (haplotypes) ตามลำดับ จากการวิเคราะห์การแบ่งกลุ่มทางพันธุกรรมโดยการสร้างเครือข่ายแฮโพลไทป์ (haplotype network) ให้ผลสอดคล้องกันเมื่อวิเคราะห์ด้วยยีน CO1 และ 16S rDNA โดยสามารถแบ่งกลุ่มพันธุกรรมของแมลงกระชอนในจังหวัดร้อยเอ็ดออกเป็น 6 กลุ่ม (I – VI) และเมื่อนำผลลำดับนิวคลีโอไทด์ของทั้งสองยีนมาวิเคราะห์เป็นแผนภูมิวิวัฒนาการ (phylogenetic tree) พบว่า ประชากรแมลงกระชอนในจังหวัดร้อยเอ็ดแบ่งสายสัมพันธ์ทางวิวัฒนาการออกเป็น 6 กลุ่ม เช่นเดียวกัน และพบค่าความแตกต่างทางพันธุกรรม p-distance เมื่อเปรียบเทียบระหว่างกลุ่มประชากรอยู่ในช่วงระหว่าง 0.0002 ถึง 0.1138 นอกจากนี้ผลการวิเคราะห์สัดส่วนของสัณฐานบางประการเปรียบเทียบระหว่างกลุ่มพันธุกรรม พบลักษณะสัณฐานที่แตกต่างกันอย่างมีนัยสำคัญ เช่น ความกว้างช่องอก ความกว้างและความยาวของปีกคู่หลัง และความยาวของปีกคู่หน้า เป็นต้น จากผลการศึกษาครั้งนี้ สามารถสรุปได้ว่า ประชากรแมลงกระชอนในแหล่งธรรมชาติของจังหวัดร้อยเอ็ดเป็นชนิดซับซ้อน (species complex) ซึ่งอาจจำแนกเป็นหลายชนิดและจำเป็นต้องศึกษาทางชีววิทยาด้านอื่นๆ รวมทั้งลักษณะทางสัณฐานทั้งภายนอกและภายในของแมลงกระชอนในกลุ่มพันธุกรรมต่างๆ อย่างละเอียดต่อไปในอนาคต เพื่อใช้เป็นข้อมูลพื้นฐานในการจัดจำแนกชนิดของแมลงกระชอนที่พบในจังหวัดร้อยเอ็ดรวมทั้งแหล่งอื่นๆ ในประเทศไทยให้ถูกต้องและชัดเจนมากยิ่งขึ้น
