Abstract: The present study evaluated the antioxidant, α-glucosidase, nitric oxide (NO) inhibitory, and cytotoxic activities of crude CH₂Cl₂, MeOH, and EtOAc extracts from the fruiting bodies of Fulvifomes fastuosus. While the CH₂Cl₂ extract was biologically inactive, further purification led to the isolation of drosophilin A methyl ether (DAME; 9) and drosophilin A (DA; 10). The EtOAc extract exhibited strong α-glucosidase and NO inhibitory activities and was further investigated phytochemically, yielding inoscavin A (2) as a major constituent, along with inoscavin E (11) and phenolic derivatives (12–14). Inoscavin A demonstrated potent α-glucosidase inhibition (IC50 = 3.22 µM), surpassing acarbose by 59-fold. These results suggest that F. fastuosus is a promising source of antidiabetic agents, with inoscavin A as a potent principle compound. The synergistic activity observed in crude extracts highlights the value of optimizing extraction strategies for therapeutic use of antidiabetic natural products.
