| dc.description.abstract | Dirty Panicle Disease presents a major constraint to rice production, causing yield losses of up to 85% during severe outbreaks, with the most noticeable symptoms emerging at harvest. Due to the limitations and environmental concerns of chemical control, biological methods have gained prominence as sustainable and eco-friendly alternatives. Recent research has focused on soil microorganisms, particularly Bacillus sp., which are known to produce antimicrobial compounds and hydrolytic enzymes that inhibit plant pathogens. This study aimed to identify native Bacillus sp. strains capable of inhibiting the fungal pathogens responsible for Dirty Panicle Disease in Khao Dawk Mali 105 rice. The research comprised five sequential experiments: The first experiment aimed to assess the pathogenicity of fungi causing Dirty Panicle Disease. This experiment evaluated disease severity and seed symptom expression caused by six fungal pathogens (Curvularia sp., Cercospora sp., Bipolaris sp., Fusarium sp., Trichoconis sp., and Sarocladium sp.) using a Completely Randomized Design (CRD) with 9 treatments and 4 replications. Seed inoculation followed ISTA standards, and symptom progression was observed at 12, 24, 48, 72, 120, and 168 hours post-inoculation. Catalase enzyme activity served as a biochemical indicator of seed deterioration. Although no external seed coat symptoms (disease level 0) were detected, internal infections significantly reduced germination. Curvularia sp. caused total inhibition (0% germination), marking it as the most aggressive, while Sarocladium sp. had the least impact (68% germination). Catalase activity significantly varied among fungi, reflecting differences in pathogenic aggressiveness. The second experiment aimed to evaluate the Bacillus sp. for antifungal activity against Dirty Panicle pathogens. Ten Bacillus isolates (UBU1, UBW2, BRCR3, BRBD12, SKKH3, SKNK4, SKKK4, SK-NK3, SKY7, and SRSHP19) were tested in cell suspension (CS) and cell-free culture filtrate (CFF) forms using a 10×6×2 factorial CRD design (120 treatments, 4 replications). Antagonistic activity was assessed using the dual culture method. The CFF of isolates SKKH3 and BRBD12 fully inhibited fungal spore germination (100%). Overall, CFF treatments were more effective than CS. PCR analysis showed that B. siamensis UBU1, BRBD12, SKKH3, SKY7, and SKKK4 carried four key antibiotic synthesis genes (srfA, ituA, bacD, and bmyA), identifying them as strong biocontrol candidates. The third experiment aimed to investigate the spore germination inhibition by selected Bacillus sp. Five high-performing isolates from Experiment 2 (B. siamensis UBU1, BRBD12, SKKH3, SKKK4, and B. Cereus SRSHP19) were tested for spore inhibition of the six fungal pathogens using a 5×6 factorial in CRD (30 treatments, 4 replications; 100 spores/treatment). Slide cultures were observed at 15 hours under a microscope. Results showed significant treatment effects. B. siamensis SKKH3 and SKKK4 exhibited the highest efficacy, completely suppressing spore germination of Curvularia sp. (100%). The fourth experiment aimed to assess the effects of Bacillus sp. on germination and seedling vigor of Khao Dawk Mali 105. To assess compatibility with seed physiology, the same five Bacillus isolates were evaluated for their effects on germination and early seedling growth in a CRD with 6 treatments and 4 replications. Surface-sterilized seeds were coated with the isolates and incubated under controlled conditions. No significant differences were observed in germination rates or germination index at 7 and 14 days. However, seedlings treated with B. siamensis SKKH3 and SKKK4 showed significantly higher vigor indices, suggesting growth-promoting effects. Importantly, none of the treatments negatively impacted germination, confirming their suitability for seed application. The fifth experiment aimed to investigate the efficacy of Bacillus sp. in reducing Dirty Panicle Disease under greenhouse conditions. This experiment tested the efficacy of the five Bacillus isolates in suppressing Dirty Panicle Disease in Khao Dawk Mali 105 under greenhouse conditions. A CRD with 38 treatments and 4 replications was used. Treatments included individual Bacillus applications with fungal inoculation, a mixed fungal inoculum, a chemical fungicide, and a commercial Bacillus product, alongside a non-inoculated control. All five Bacillus isolates significantly reduced disease incidence compared to the untreated control. They also improved 1,000-grain weight, weight of unbroken grains, and total yield per plot. These parameters were significantly better than the distilled water control but not significantly different from chemical or commercial Bacillus-based treatments. This study demonstrates that native Bacillus sp., particularly B. siamensis SKKH3 and SKKK4, exhibit strong potential for suppressing fungal pathogens associated with Dirty Panicle Disease. The recommendation is to develop the product into a bioformulation for actual commercial use and to find ways to create a formulation with high stability and long shelf life, offering a safe and sustainable alternative for the production of Khao Dawk Mali 105 rice under varying environmental conditions across different regions. | en |
